Title: The Integrated Stress Response Regulates Pain Hypersensitivity

The integrated stress response (ISR) is an adaptive response to various cellular stressors, including unfolded proteins, viral infection, and inflammation. The ISR acts as a central mechanism that underlies pain hypersensitivity in diabetes and Multiple Sclerosis (MS). Methylglyoxal (MGO) is a reactive glycolytic metabolite elevated in the plasma of patients suffering from neuropathic pain. By investigating how MGO triggers ISR, I aim to uncover the role of translational control mechanisms, particularly those regulated by eIF2A, that cause nociceptor hyperexcitability and pain. Using techniques like TRAP-seq and human tissue models, I will identify molecular targets to improve the understanding of diabetic neuropathy and MS with the goal of clinical translation. I will also explore innovative, non-opioid analgesics, such as TMEM97 modulators, to inhibit ISR and develop effective treatments for diabetic and MS neuropathic pain, addressing significant unmet clinical needs.
