Title: Unlocking the Mammalian Spinal Dorsal Horn: Insights Into Key Circuits and Novel Cross-Species Research Tools

The spinal dorsal horn serves as a critical hub for integrating somatosensory information, including acute pain, temperature, and itch, ultimately relaying this data to the brain to form sensory perceptions. Our laboratory has been diligently investigating the neural circuitry the encodes mechanical allodynia—a condition in which normally innocuous mechanical stimuli are perceived as painful following an injury. Utilizing mice as a model system, we have identified key components of the mechanical allodynia circuitry, leading to the significant discovery that distinct types of injuries—both inflammatory and neuropathic—recruit overlapping yet unique neural circuits. In this seminar, we will present new data elucidating the mechanisms by which mechanical allodynia induced by polyneuropathic injuries is encoded. We will provide compelling evidence that highlights the crucial role of a specific population of neurons, traditionally recognized for their exclusive association with itch. Additionally, we will introduce a novel species integrated definition of neuron subtypes within the dorsal horn and outline a strategic plan that will enable researchers genetic access to these neuron subtypes across a range of species, including both mice and humans. This presentation promises to offer valuable insights into the complexities of pain mechanisms and new tools for future research advancements.
